Normal role description: | Transcription factor that can act as activator and suppressor. Binds to its own promotor in a positive feedback loop. The recognition sequence is 5'-CACCC-3'. KLF4 inhibits cell cycle by regulating the expression of cell cycle control proteins. Mediates between Notch and Wnt signalling pathways. Involved in differentiation of epithelial cells, but prevents embryonic stem cell differentiation. It was shown to regulate the function of p53 and suggested to be involved in decreased p53 transcription. In response to DNA damage it promotos p21 expression- a checkpoint protein, which inhibits cell cyle progression. Expression levels are decreased in colorectal and gastric tumors, but elevated in breast cancer and oral squamous-cell carcinomas, suggesting that it acts as an oncogene in these tissues. |
