Normal role description: ENO1 is a multifunctional enzyme that is involved in glycolysis and various other processes such as growth control, tolerance to hypoxia and allergic responses by stimulating immunoglobulin production. It may serve as a receptor and activator of plasminogen on the cell surface of for example leukocytes and neurons. The gene is upregulated in numerous cancer, but alternative splicing of the gene resulting in a shorter isoform (MBP-1) located in the nucleus, has been shown to bind to the c-Myc promoter and function as a tumour suppressor.
Commentary on involvement of protein in cancer: 25% (1/4) of upper aerodigestive tract samples (pharynx) were also mutated (mis-sense), and so was 1/101 kidney- and 1/468 CNS samples. Only 2 ovarian samples were included (Sanger COSMIC). The chromosome section where the gene is located is often deleted in human malignancies, including neuroblastoma, melanoma, pheochromocytoma, breast, liver and colon cancer.
