Cancer Protein Description

This report provides a detailed description of a selected cancer protein with information collected from various sources, including UniProt, the Wellcome Trust Sanger Institute’s Catalogue of Somatic Mutations in Cancer (COSMIC), and the Atlas of Genetics and Cytogenetics in Oncology and Haematology.

Protein Name: TPR
Gene Name: TPR
Protein Full Name: Nucleoprotein TPR
Alias: Nucleoprotein TPR; Translocated promoter region; Translocated promoter region (to activated MET oncogene)
Mass (Da): 267293
Number AA: 2363
UniProt ID: P12270; Q15624
Locus ID: 7175
COSMIC ID: TPR
Gene location on chromosome: 1q25*
Cancer protein type: OP
Effect of cancer mutation on protein: GAIN
Effect of active protein on cancer: PROMOTES
Number of cancer specimens: 19805
Percent of cancer specimens with mutations: 1.67
Deregulated in translocations: Translocation --> TPR-MET in gastric cancer; and TRK-T1 (TPR-NTRK1) in thyroid papillary carcinoma
Normal role description: TPR is a coiled-coil protein that forms intranuclear filaments attached to the inner surface of nuclear pore complexes, and interacts directly with several components of the complex. It is required for the nuclear export of mRNAs and some proteins. Oncogenic fusions of the 5' end of the gene with several different kinase genes have been observed in some neoplasias such as thyroid papillary carcinoma and gastric cancer.
Commentary on involvement of protein in cancer: 1/45 CNS tumour samples was also mutated. Only 1 liver-, 2 ovarian, and 2 bladder tumour samples were included in Sanger COSMIC. *Note: Chromosome location = 1q31.1 according to OMIM
