Normal role description: Links activated growth factor receptors with signalling pathways. The isoforms p46Shc and p52Shc, connect receptor tyrosine kinases with Ras via Grb2/Sos. The isoform p66Shc is a p53 target and is crucial for induction of apoptosis. May regulate life span and effects of ROS. p52/p46Shc is indirectly associated with cell proliferation and carcinogenesis (overexpression or hypermethylation in many tumour types where receptor-tyrosine kinases are constitutively activated), whereas the function of p66 in tumorigenesis remains controversial.
