Normal role description: | SPAM1 is a GPI-anchored enzyme that is located on the human sperm surface and inner acrosomal membrane. It functions to degrade hyaluronic acid (HA) -- a structural proteoglycan found in extracellular matrices (ECM) and basement membranes. Dereguation or aberrant SPAM1 has been implicated in tumour progression, as increased degradation of the ECM by SPAM1 effectively weakens a critical barrier -- the basement membrane -- thereby aiding tumour metastisis. Mutations in this gene have been observed in upper aerodigestive tract (mouth and larynx) and ovarian tumors. Upregulation of this gene is associated with breast, laryngeal, colon, and skin cancer. |
