Normal role description: | GRB10 is an adapter protein and a growth factor receptor-binding protein that interacts with insulin receptors and insulin-like growth-factor receptors. Overexpression of some isoforms of the protein will lead to inhibition of tyrosine kinase activity and results in supression of growth. It exerts its regulatory effects by inhibiting signalling pathways and by increasing receptor degradation. It is responsible for delaying and reducing AKT1 phosphorylation in response to insulin stimulation and for blocking binding of INSR and IRS1 and IRS2, and for preventing insulin-stimulated IRS1 and IRS2 tyrosine phosphorylation. It recruits NEDD4 to IGF1R, leading to ubiquitination of the receptor and increased proteasomal degradation. It negatively regulates Wnt signalling, and is a positive regulator of the KDR/VEGFR-2 signalling pathway. Expression of the gene observed in the brain are from the paternal allele whereas the gene expression in the placental trophoblasts is from the maternal allele. |
