Normal role description: | De-acetylates many protein targets. SIRT1 contains nuclear localization signal but also functions to regulate metabolic processes in the cytoplasm. SIRT1 is implicated in histone de-acetylation and therefore activation/repression of different genes. For example, it protects against p53-dependent apoptosis but also functions as a cell-cycle checkpoint protein via FOXO1 and FOXO4 interaction; inhibits NF-kB signalling pathways. These varied roles lead to both tumour promotion and inhibition in different cancer systems |
