Normal role description: | SCN3B is the beta subunit for voltage gated sodium channels. Sodium channels consist of a pore forming alpha subunit which associate with one to two beta subunits responsible for regulating channel gating as well as promoting cell adhesion. The SCN3B isoform confers slower kinetics of channel opening than the SCN1B isoform. SCN3B may have a role in p53 mediated apoptosis. p53 tumour suppressor protein was found to upregulate SCN3B expression in cell lines exposed to DNA damaging agents. Localization of SCN3B was at the ER instead of the plasma membrane when exposed to DNA damaging agents. Trangenic introduction of SCN3B into transformed cells could suppress colony formation and induce apoptosis. Thus SCN3B may act as a tumour suppressor protein, however, the exact mechanism through which it exerts its effects is still unknown but is speculated to occur through ER stress response pathways. |
