Normal role description: TRIM33 acts as an E3 ubiquitin-protein ligase that promotes SMAD4 ubiquitination, nuclear exclusion and degradation via the ubiquitin proteasome pathway. It may also act as a transcriptional repressor by inhibiting the transcriptional response to the TGFβ/BMP signalling cascade. Furthermore, it plays a role in the control of cell proliferation, and stimulates erythroid differentiation of hematopoietic stem/progenitor cells via its association with SMAD2 and SMAD3.
Commentary on involvement of protein in cancer: *Note; translocations of the gene generates a fusion oncoprotein (GAIN of function), but the TRIM33 has also been found to function as a TSP in human cancers such as hematopoietic malignancies. 1/257 breast-, 3/477 CNS-, 1/42 colorectal-, 3/171 lung-, an 1/85 ovarian tumour samples were also mutated (Sanger COSMIC)
