Cancer Protein Description

This report provides a detailed description of a selected cancer protein with information collected from various sources, including UniProt, the Wellcome Trust Sanger Institute’s Catalogue of Somatic Mutations in Cancer (COSMIC), and the Atlas of Genetics and Cytogenetics in Oncology and Haematology.

Protein Name: NEU3
Gene Name: NEU3
Protein Full Name: Sialidase-3
Alias: Ganglioside sialidase; Membrane sialidase; N-acetyl-alpha-neuraminidase 3; NEUR3; Sialidase 3
Mass (Da): 48252
Number AA: 428
UniProt ID: Q9UQ49
Locus ID: 10825
COSMIC ID: NEU3
Gene location on chromosome: 11q13.4
Cancer protein type: OP
Effect of cancer mutation on protein: UNCLEAR
Effect of active protein on cancer: PROMOTES
Number of cancer specimens: 19557
Percent of cancer specimens with mutations: 0.31
Normal role description: NEU3 is a glycohydrolytic enzyme localized in the plasma membrane responsible for removing sialic acid moieties of gangliosides, and thereby altering ganglioside composition of tissues and cells. It has also been shown to interact with signalling molecules such as EGFR, Grb2 and Rac, and to be modulating for example EGFR and IR signalling. Over-expression of the gene has been linked to cancer progression in colon- and renal cell cancer, and has been implicated in Diabetes Mellitus type II.
Commentary on involvement of protein in cancer: Only 2 ovarian cancer samples were included in Sanger COSMIC. 1/1 pharyngeal cancer sample was also mutated (non-sense).
